How to Use a Drug Test Cup — Step-by-Step Guide

A simple step-by-step procedure for administering a rapid urine drug test cup. Total time: about 5-7 minutes from collection to readable result.

What you'll need

Supplies:

  • Sealed drug test cup (room temperature)
  • Disposable gloves
  • Clean collection area
  • Documentation/chain-of-custody form (if applicable)

Tools:

  • Timer or stopwatch
  • Pen for marking results

Step-by-step procedure

Step 1: Verify the test is unexpired and at room temperature

Check the expiration date printed on the foil pouch. Tests stored below 36°F or above 86°F should be discarded. Cold tests must be brought to room temperature (15-30 minutes) before opening.

Step 2: Open the pouch and inspect the cup

Tear the foil pouch open along the notch. Remove the cup and verify the integrity check strip is intact. Do NOT open the lid yet — keep the cup sealed during specimen collection.

Step 3: Collect the urine specimen

Have the donor urinate directly into the cup, filling to the level indicator line (typically 30-45 mL). Replace the cap immediately and tighten securely.

Step 4: Check specimen temperature within 4 minutes

Look at the temperature strip on the side of the cup. A valid specimen should read 90-100°F (32-38°C) within 4 minutes of collection. Out-of-range temperatures may indicate specimen tampering.

Step 5: Activate the test (if applicable)

Most rapid cups are self-activating once the urine contacts the test strips. Some models require you to remove a peel tab on the side of the cup to expose the result window.

Step 6: Wait for the result to develop

Set a timer for 5 minutes. The control line (C) appears first to confirm the test is working. Test lines (T) appear next to each drug abbreviation indicating a NEGATIVE result. Missing test lines indicate PRESUMPTIVE POSITIVE.

Step 7: Read the result

Within the 5-10 minute window: a visible C line plus visible T line = negative for that substance. A visible C line plus NO T line = presumptive positive. NO C line = invalid test, retest with a new device.

Step 8: Document and confirm if positive

Record results on your chain-of-custody form. For employment or legal use, send any positive specimen to a SAMHSA-certified laboratory for GC-MS or LC-MS/MS confirmation. Never act on a screening result alone.
